Background

GAS6 Protein is encoded by the growth arrest-specific gene 6 (Gas6) and belongs to the vitamin K-dependent protein family[2].
GAS6 Protein-AXL can regulate various signaling pathways like PI3K/AKT/mTOR, JAK/STAT, NF-κB, and RAS/RAF/MEK/ERK. It plays a role in tumor cell survival, anti-apoptotic signaling, mitosis, migration, invasion, drug resistance, angiogenesis, and the tumor-host relationship[2].
GAS6 Protein-AXL can promote the survival of endothelial cells, regulate the optimal cytokine signaling during the development of human natural killer cells, support liver regeneration, assist the survival and migration of gonadotropin-releasing hormone neurons, and facilitate platelet activation or thrombotic responses; it can also induce actin remodeling, driving membrane folding, giant cell phagocytosis, and cancer cell invasion[1][3].
GAS6 Protein signaling plays a beneficial role in septic heart conditions by inhibiting inflammation, oxidative stress, mitochondrial damage, and ER stress, with NLRP3 being the main molecule mediating GAS6 signaling[4].
GAS6 Protein mediates the binding of viruses to target cells by bridging the phosphatidylserine on the viral particle to the TAM receptor tyrosine kinase AxI on the target cell, facilitating viral entry[5].
GAS6 Protein is closely related to pathological processes like nephritis, tumors, viral infections, and cardiovascular diseases[1][4][5].

In Vitro

GAS6 Protein can trigger membrane folding in human glioblastoma LN229 cells, manifested as the formation of peripheral folds (PR) and circular dorsal folds (CDR)[1].
GAS6 Protein (25-100 ng/mL, 3 h) can inhibit the expression of inflammatory factors and NLRP3, ASC proteins, thereby reducing LPS induced inflammation in HL-1 cells[4].

In Vivo

Loss of GAS6 Protein expression in GAS6 knockout (GAS6 KO) mice leads to exacerbation of septic myocardial injury in septic mice[4].
GAS6 Protein (2 μg; once daily; 10 days; i.p.) induces erythropoiesis and alleviates anemia in GAS6-/-anemic mice[6].
GAS6 Protein (2 μg; once daily; 4 days; injection) can increase proteinuria and the number of PCNA positive cells in each glomerulus of GAS6-/-nephrotoxic nephritis mice, thereby promoting the development of nephritis[7].

Biological Activity

Immobilized Human AXL-His at 5 μg/mL (100 μl/well) can bind Human GAS6-Fc. The ED50 of Human GAS6-Fc is ≤27 μg/mL.
