Background

HGFR protein is a receptor tyrosine kinase that transduces signals from the extracellular matrix to the cytoplasm through its binding to hepatocyte growth factor/HGF ligand. This interaction regulates various physiological processes, including proliferation, scattering, morphogenesis, and cell survival. Upon ligand binding, HGFR undergoes autophosphorylation on its intracellular domain, providing docking sites for downstream signaling molecules such as PIK3R1, PLCG1, SRC, GRB2, STAT3, and GAB1. Activation of HGFR leads to the activation of multiple signaling cascades, including RAS-ERK, PI3 kinase-AKT, and PLCgamma-PKC. These signaling pathways are associated with morphogenetic effects, prosurvival effects, and the regulation of various developmental processes, such as gastrulation, neuronal precursor migration, angiogenesis, kidney formation, and skeletal muscle development. In adults, HGFR is involved in wound healing, organ regeneration, tissue remodeling, and the differentiation and proliferation of hematopoietic cells. Additionally, HGFR acts as a receptor for the internalin InlB of Listeria monocytogenes, enabling the entry of this pathogen into host cells. Moreover, HGFR may play a role in cortical bone osteogenesis.
