MCP-5/CCL12 protein, Mouse (CHO) is a potent monocyte-active chemokine that binds to the CCR2 chemokine receptor and is involved in allergic inflammation and host responses to pathogens. MCP-5/CCL12 protein, Mouse (CHO) is a recombinant mouse MCP-5/CCL12 (G23-G104) expressed by CHO[1].

Background

CCL12, also known as monocyte chemotactic protein 5 (MCP-5), belongs to the CC chemokine cluster on mouse chromosome 11. CCL12 is a potent peripheral blood mononuclear cell chemokine that attracts eosinophils, monocytes and lymphocytes but not neutrophils. It induces calcium flux in peripheral blood mononuclear cells, but not in purified mouse eosinophils or neutrophils. CCL12 is a potent monocyte-active chemokine that signals through CCR2. Involved in allergic inflammation and host response to pathogens, and may play a key role in the early stages of allergic pulmonary inflammation[1][2].

In Vivo

CCL12 (50 ng/mL) can induce fibroblast migration in wild-type Balb/c mice. However, fibroblasts from CCR2-/- mice are unable to migrate to CCL12. CCL12 expression increases at day 1 after fluorescein isothiocyanate (FITC)-induced fibrosis, peaks at day 3 after FITC, and remains elevated until day 21. And neutralization of CCL12 in wild-type mice significantly prevents FITC-induced fibrosis[3].

Biological Activity

The ED50 is <0.5 μg/mL as measured by CHO-K1/Gα15/mCCR2 cells (human Gα15 and mouse CCR2 stably expressed in CHO-K1 cells).
