The preproprotein undergoes proteolytic processing by plasmin or trypsin, generating heavy and light chains that associate via disulfide linkages to form the heterodimeric enzyme. T-PA's enzymatic activity plays a pivotal role in maintaining the delicate balance between fibrinolysis and coagulation. Increased activity leads to hyperfibrinolysis, resulting in excessive bleeding, while decreased activity can lead to hypofibrinolysis, potentially causing thrombosis or embolism. Alternative splicing produces multiple transcript variants, including at least one isoform that undergoes proteolytic processing. The gene exhibits broad expression, with notable levels observed in the urinary bladder (RPKM 61.1), placenta (RPKM 41.3), and 18 other tissues, reflecting its diverse roles in various physiological contexts.
