Background

The YWHAH protein functions as an adapter implicated in the regulation of a broad spectrum of both general and specialized signaling pathways, recognizing phosphoserine or phosphothreonine motifs to bind with numerous partners. This binding typically leads to the modulation of the activity of the interacting partner. YWHAH negatively regulates the kinase activity of PDPK1 and exists as a homodimer. It interacts with various nuclear hormone receptors and cofactors, including AR, ESR1, ESR2, MC2R, NR3C1, NRIP1, PPARBP, and THRA. Additionally, it interacts with ABL1 in its phosphorylated form, retaining it in the cytoplasm, and weakly interacts with CDKN1B. Other interactions involve ARHGEF28, CDK16, GAB2, KCNK18 (in a phosphorylation-dependent manner), SAMSN1, the 'Ser-241' phosphorylated form of PDPK1, the 'Thr-369' phosphorylated form of DAPK2, PI4KB, TBC1D22A, TBC1D22B, SLITRK1, and MEFV.
