Description

Secalciferol ((24R)-24,25-Dihydroxyvitamin D3) is the major active metabolite of Vitamin D. Secalciferol activates vitamin D receptor (VDR) with an EC50 value of 150 nM. Secalciferol is involved in a wide range of biological functions such as calcium homeostasis, cellular differentiation and proliferation processes, as well as other functions related to the immune system, which is promising for research of rickets, osteomalacia, hypercalcemia and autoimmune disorders[1][2][3][4][5].

In Vitro

Secalciferol of a part of the glucuronides is deconjugated by the enzyme from bacteria in the intestine and is probably reabsorbed and further oxidized to other metabolites in rat intestine[2].
Secalciferol is metabolized by β-glucuronidase to 24,25(OH)2D3-3G and -24G in rat bile[2].
Secalciferol (100 pM-10 μM, 24 h) dose-dependently activates vitamin D receptor (VDR) with an EC50 value of 150 nM, similar to that by Calcitriol (HY-10002) in the IZ-CYP24 and 12-VDRE cells[3].

In Vivo

Secalciferol (1-1000 μg/kg, i.p., daily for 28 days) dose dependently increases the length of the fifth lumbar spine in the hypophosphatemic (Hyp) mice[1].
Secalciferol (1 mg/kg, s.c., daily for 28 days) improves the rachitic bone and increases mineralized bone volume in the Hyp mice[1].
Secalciferol (0 or 5 ppm, powder fiet for 57 weeks) induces hyperplasia and neoplasia of the adrenal medulla with increase in cell prpliferative activity in rats[4].
