Maytansinoids

Maytansinoids (DMs) interfere with microtubule assembly but are mechanistically similar to vinca alkaloids. Maytansines take effect by binding and capping the ‘plus’ end of the growing microtubule and block the polymerization of tubulin dimers preventing the formation of mature microtubules, causing mitotic arrest. Of the two DMs, DM1 and DM4 are widely used as ADC payloads.
