Description

Collagen, Type II, from bovine snout membrane is a type II collagen that can induce osteoarthritis by eliciting a strong and sustained T cell response. Type II collagen can be used to model collagen-induced arthritis in mice[1].

Background
Collagen, Type II, from bovine snout membrane induces arthritis by triggering a strong and sustained T-cell response.
Specific Mmodeling Methods
Mice: DBA/1 • male • 10-12 weeks old
Administration: 200 μg • immunised intradermally in two sites at the base of the tail
Note
(1) All of this collagen is in CFA. To prepare CFA: grind 100 mg of dried inactivated Mycobacterium tuberculosis H37Ra into a fine powder using a mortar and pestle, then suspend it in 30 mL of incomplete Freund's adjuvant.
(2) Fights between male mice reduce the incidence of arthritis. To minimize the risk of fighting, mice older than 6 weeks aren't mixed together in different cages.
(3) Within 2 weeks after the onset of illness (early arthritis) or within 6-8 weeks after the onset of illness (late arthritis), euthanize the arthritis mice, decalcify the joints, embed in paraffin, and conduct histological evaluation.
Modeling Indicators
Molecular changes: Generate strong and sustained T cell responses.
Organizational changes: Inflammatory infiltration and destruction of joints, with a large accumulation of polymorphonuclear cells (PMNs).
Phenotypic observation: Swelling of the claws occurs.
