Description

Chrysene is a high molecular weight (HMW), polycyclic aromatic hydrocarbon (PAH) known for its recalcitrance and carcinogenic properties[1].

Chrysene can be used in animal modeling to create hepatic tumor models.

Induction of liver tumors[2][3][4][5]
Background
Chrysene exerts its carcinogenic effects by being metabolized by monooxygenases into carcinogenic metabolites (epoxides and their hydrolysis products). Additionally, studies have shown that chrysene may induce oxidative liver damage, including liver DNA damage and glutathione depletion, by activating the aryl hydrocarbon receptor (AhR), which may be related to its carcinogenic risk[2][3][4][5]
.
Specific Mmodeling Methods
Mice: CD-1 • male • 1, 8, and 15 days old
Administration: 700 or 2800 nmol/Mouse • ip • administered on postnatal days 1, 8, and 15
Note
(1) Chrysene is dissolved in DMSO, and the total amount of Chrysene injected intraperitoneally into each mouse is either 700 or 2800 nmol. The distribution of the three injection doses is as follows:
On Day 1, inject 100 or 400 nmol of Chrysene, with a DMSO volume of 10 μL.
On Day 8, inject 200 or 800 nmol of Chrysene, with a DMSO volume of 20 μL.
On Day 15, inject 400 or 1600 nmol of Chrysene, with a DMSO volume of 40 μL.
(2) The administration is performed under dim light, and the mice are euthanized one year later to determine the incidence of liver tumors.
