Description

NBI-31772 is a non-selective IGFBP inhibitor (Ki=47 nM). NBI-31772 has neuroprotective effects and reduces infarct volume during cerebral ischemia. NBI-31772 can also restore or enhance proteoglycan synthesis in osteoarticular chondrocytes. In addition, NBI-31772 also increases the effect of IGF3 on zebrafish oocyte maturation[1][2][3][4].

In Vitro

NBI-31772 (0.1-10 µM; 24 h) stimulates the synthesis of IGF-1-dependent proteoglycans in human OA chondrocytes in a dose-dependent manner[3].
NBI-31772 (0.1-10 µM; 24 h) significantly stimulates the percentage of rabbit articular chondrocyte-associated proteoglycans in the presence of IGF-1 and IGFBP-3[3].
NBI-31772 (5-100 µM; 16 h) dose-dependently induces the maturation of zebrafish oocytes[4].

In Vivo

NBI-31772 (5-100 μg; siv; administered at the onset of ischemia) dose-dependently reduces total infarct volume and cortical infarct volume at the onset of ischemia in rats with middle cerebral artery occlusion (MCAO) model[2].
NBI-31772 (50 μg; i.v.; administered 0, 1, 2, or 3 hours after MCAO) reduces cortical infarct size and brain swelling in rats with intraluminal suture MCAO model[2].
NBI-31772 (0.4 μM/strip; i.p.; 4 h) induces germinal vesicle rupture in zebrafish[4].
