Description

NOD1 antagonist-2 (compound 66) is an orally active selective antagonist of both human and mouse NOD1. NOD1 antagonist-2 (compound 66) antagonizes NOD1-induced NF-κB and MAPK pathways[1].

IC50 & Target[1]

hNOD1

0.8 nM (IC50, in HEK293 cells)

mNOD1

32 nM (IC50, in HEK293 cells)

In Vitro

NOD1 antagonist-2 (compound 66) demonstrates promising antagonistic activity against both h/mNOD1-induced NF-κB activation in HEK293 cells (IC50 = 0.8 nM and 32 nM, respectively)[1].
NOD1 antagonist-2 (compound 66, 0.1, 1 μM) markedly inhibits the increases in p-RIP2, p-p65, p-p38, and p-JNK of both THP-1 cells and mBMDMs, demonstrating its efficacy in modulating NOD1-induced signaling pathways[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

In Vivo

NOD1 antagonist-2 (compound 66, orally) inhibits the expression of inflammatory cytokines (IL-6, TNF-α) and chemokines (CXCL1, CXCL8) in immune and epithelial cells, as well as inflammatory cytokines (KC, IL-6) in a H. pylori-induced murine model[1].

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
