nano-Differential Scanning Fluorimetry (nanoDSF) is a powerful biophysical technique for studying the stability and folding properties of proteins and other biomolecules. This method utilizes the intrinsic fluorescence of tryptophan and tyrosine residues in proteins and the changes in fluorescence signals to provide insights into protein conformational changes under varying temperature and chemical conditions.

How nanoDSF Works

Tryptophan and tyrosine residues in proteins can generate fluorescent signals, and nanoDSF technology can determine the melting temperature (Tm) by monitoring changes in protein conformation during the heating process through endogenous fluorescence signals. When ligand binding enhances protein stability, the melting temperature (Tm)—the temperature at which the amounts of folded and unfolded protein are equal—will also increase. Therefore, this method can be utilized for rapid verification of protein-ligand binding activity and for high-throughput qualitative screening of potential ligands.
