It features high sensitivity and does not require labeling.

How ITC Works

ITC operates by titrating a solution with a ligand (small molecule) into a titrant containing a target macromolecule (such as a protein or nucleic acid) within a calorimeter. As the ligand is added, the binding event occurs, resulting in either the release or absorption of heat. The microcalorimeter constantly measures these heat changes, which are plotted as a titration curve. The area under the peaks in the curve correlates to the amount of heat released or absorbed, allowing for precise calculations of various thermodynamic parameters. Information such as affinity (KD), stoichiometric ratio (n), enthalpy change (ΔH) and entropy change (ΔS) of molecular interactions can be obtained through ITC experiments.
