    Basement Membrane Matrix HC (Phenol Red) (HY-K6005) (HY-K6005:HCT-116 cell suspension = 1:1; s.c. a single dose) promotes the tumor formation (HCT-116 subcutaneous xenograft) in 4-5 week old BALB/c-nu nude mice.

    MCE Basement Membrane Matrix HC (Phenol Red) is a natural basement membrane matrix extracted from mouse tumors and is composed mainly of various growth factors and extracellular matrix components. The main extracellular matrix components are: Laminin, Col-IV, Entactin, Heparan sulphate proteoglycans, etc. The main growth factor components are: insulin-like growth factor (IGF-1), transforming growth factor beta (TGF-β), vascular endothelial growth factor (VEGF), epidermal growth factor (EGF), fibroblast growth factor (bFGF), etc. The percentage of each of these growth factors is higher than that of standard basement membrane matrix.

    MCE Basement Membrane Matrix HC (Phenol Red) is mainly used for angiogenesis experiments, animal models and 3D tumor model construction.

    Storage

    -20°C, 2 years.

    Avoid repeated freezing and thawing.

    Protocol

    a. Use a pre-cooled pipette to blow the matrix gel evenly after thawing.

    b. Dilute MCE Basement Membrane Matrix HC with cell suspension at 4°C in a 1:1 ratio to a suitable final concentration.

    c. Inject diluted cell suspension at a depth of 1 cm under the skin on nude mice. The cell suspension should be injected in 30 minutes and be placed on ice to slow down apoptosis and prevent from gelling.

    d. Formed tumors can be seen in about 1 week.

     

    Note: When thawing the product for the first time, immerse the vial in crushed ice and place it in a 4°C to melt. After melting, mix thoroughly before aliquoting, and then store at -20°C.

    • Do most proteins show cross-species activity?

      Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
