Noggin Protein, Human (HEK293) is a recombinant Noggin protein produced by HEK293 cells.

Background

Noggin Protein belongs to the bone morphogenetic protein (BMP) antagonist family of the TGF-β superfamily. Noggin Protein is a glycosylated cysteine knot chemokine protein. Noggin Protein binds to BMP to form a neutralizing complex, preventing BMP from binding to the receptor, thereby inhibiting BMP signaling and affecting cell survival, proliferation and differentiation[1]. Noggin Protein can be expressed in human arterial endothelial cells and bone marrow mesenchymal stem cells[1]. Noggin Protein antagonizes BMP4-induced hypertension, inhibits angiogenesis, regulates mesenchymal stem cell differentiation, and maintains embryonic stem cell pluripotency[1][2][3][4][5]. Noggin Protein, Human is composed of 232 amino acids, forming a covalently linked homodimer, and has the ability to bind to BMP4 with high affinity.

In Vitro

Noggin Protein, Human (200 ng/mL; 3-21 days) induces alkaline phosphatase activity and mineralization alone or synergistically with inflammatory cytokines/Dexamethasone (HY-14648) in human mesenchymal stem cells (HMSCs), upregulating BMP-2 and ActRII gene expression[2].

In Vivo

Noggin Protein, Human (0.45-0.9 mg/kg; subcutaneous osmotic pump implantation; 4 weeks) antagonizes BMP4-induced hypertension, vascular NADPH oxidase activation, and endothelial dysfunction in C57BL/6 and apoE-/- mice[5].

Biological Activity

1.Measured by its ability to inhibit BMP-4-induced alkaline phosphatase production by ATDC5 mouse chondrogenic cells. The IC50 is < 1.97 ng/mL, corresponding to a specific activity of > 5.08×105 units/ mg.
2.Measured by its ability to inhibit BMP-2-induced alkaline phosphatase production by ATDC5 mouse chondrogenic cells The ED50 for this effect is <2 μg/mL in the presence of 2000 ng/mL of Recombinant Human BMP-2.
3.Measured by its ability to inhibit BMP-4-induced alkaline phosphatase production by ATDC5 mouse chondrogenic cells. The ED50 for this effect is <6.0 ng/mL in the presence of 50 ng/mL of Recombinant Human BMP-4, corresponding to a specific activity is >2.0×105 units/mg.
4.Measured by its ability to inhibit BMP-4-induced alkaline phosphatase production by ATDC5 mouse chondrogenic cells. The ED50 for this effect is <3.0 ng/mL in the presence of 30 ng/mL of Recombinant Human BMP-4, corresponding to a specific activity is >3.33×105 units/mg.
5.Measured by its ability to inhibit BMP-4 induced alkaline phosphatase production by ATDC5 mouse chondrogenic cells. The ED50 for this effect is 10-100ng/mL in the presence of 200 ng/mL of Recombinant Human BMP 4.

  • Measured by its ability to inhibit BMP-4-induced alkaline phosphatase production by ATDC5 mouse chondrogenic cells. The IC50 is 1.523 ng/mL, corresponding to a specific activity of 6.6×105 units/ mg.
