Description

Anamorelin (RC-1291) hydrochloride is an orally active Ghrelin receptor agonist with an EC50 of 0.74 nM. Anamorelin hydrochloride can promote appetite, increase body weight, and stimulate the secretion of growth hormone and insulin-like growth factor-1. Anamorelin hydrochloride can be used in the research of anorexia and cancer cachexia[1][2].

In Vitro

Anamorelin (0.01 nM-10 μM; 15 min) hydrochloride stimulates growth hormone release in a dose-dependent manner in rat pituitary cells, with an EC50 of 1.5 nM[1].

Anamorelin (3-30 mg/kg; oral administration; 6 days) hydrochloride increases food intake and body weight in rats in a dose-dependent manner[1].
Anamorelin (3.5 mg/kg; oral administration; 1-7 days) hydrochloride significantly elevates the levels of growth hormone and insulin-like growth factor-1 in pigs[1].
Anamorelin (3-30 mg/kg; oral administration; 28 days) hydrochloride does not promote tumor growth in a nude mouse model of lung cancer xenografts, but increases the body weight of nude mice[2].

Animal Model: Female athymic nude mice (nu/nu, Harlan) (18.4-26.6 g, 9-10 weeks old) treated A549 cells[2]
Dosage: 3, 10 and 30 mg/kg

Result: Did not promote tumor growth in the murine NSCLC xenograft model.
Significantly increased the percentage of mean body weight gain at the end of treatment at doses of 10 and 30 mg/kg compared with controls.

Select the appropriate dissolution method based on your experimental animal and administration route.

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for in vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

  • Protocol 1

    Add each solvent one by one:  10% DMSO    90% Corn Oil

    Solubility: ≥ 5 mg/mL (8.57 mM); Clear solution

    This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown). If the continuous dosing period exceeds half a month, please choose this protocol carefully.

    Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 900 μL Corn oil, and mix evenly.

  • Protocol 2

    Add each solvent one by one:  10% DMSO    40% PEG300    5% Tween-80    45% Saline

    Solubility: ≥ 2.5 mg/mL (4.29 mM); Clear solution

    This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
