Description

Palmitoylcarnitine chloride is a fatty acid-derived mitochondrial substrate, and selectively decreases cell survival in colorectal and prostate cancer cells by affecting on pro-inflammatory pathways, Ca2+ influx, and DHT-like effects[1].

In Vitro

Palmitoylcarnitine (5-20 μM, 5 min) increases [Ca2+]i in a dose-dependent manner in adult rat cardiomyocytes, with 15 μM showing a significant and reversible effect without affecting cell viability[2].
Palmitoylcarnitine (15 μM, 5 min) increases [Ca2+]i in adult rat cardiomyocytes through L-type Ca2+ channels, Na+-Ca2+ exchange, and sarcoplasmic reticulum Ca2+ release[2].
Palmitoylcarnitine (5, 10 μM, overnight) inhibits Akt Ser473 phosphorylation and reduces insulin signaling in C2C12 cells[3].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Western Blot Analysis[2]

Cell Line: C2C12 myoblasts (mouse skeletal muscle)
Concentration: 5, 10 μM
Incubation Time: overnight incubation (12 h)
Result: Reduced insulin-stimulated Akt Ser473 phosphorylation.
In Vivo

Palmitoylcarnitine (10 mg/kg, s.c., once daily for 28 days) induces insulin resistance and hyperinsulinemia, leading to glucose metabolism impairment in CD-1 mice[3].
Palmitoylcarnitine (50 mg/kg, s.c., single dose) reduces insulin sensitivity and induces glucose intolerance and metabolic dysfunction in CD-1 mice[3].
Palmitoylcarnitine (50 mg/kg, i.p., single dose) increases muscle long-chain acylcarnitine content and induces insulin resistance in CD-1 mice, characterized by hyperglycemia and reduced muscle glucose uptake[3].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: Palmitoylcarnitine (HY-126357)-induced insulin resistance CD-1 mouse model[3]
Dosage: 50 mg/kg (s.c. or i.p., single dose) or 10 mg/kg (s.c., long-term administration)
Administration: Intraperitoneal injection (i.p.), single dose;
Continuously released for 24 hours via a subcutaneous osmotic minipump (s.c.)., single dose
Subcutaneous osmotic minipump (s.c.), once daily for 28 days.
Result: Increased muscle long-chain acylcarnitine content, elevated blood glucose levels in both fasted and fed states, and decreased muscle glucose uptake (i.p.).
Induced glucose intolerance, delayed energy metabolism shift, and reduced insulin sensitivity (s.c., single dose).
induced insulin resistance and hyperinsulinemia, blocked insulin-mediated blood glucose reduction, increased blood lactate levels, and impaired glucose metabolism (s.c., long-term administration).

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
