17-phenyl trinor PGF2α N-ethyl amide is an F-series prostaglandin analog which has been approved for use as an ocular hypotensive drug, sold under the Allergan trade name Bimatoprost.1 The N-ethyl amide prostaglandin prodrugs are converted to the active free acid more slowly than the analogous prostaglandin ester prodrugs such as latanoprost.2 This product is the isopropyl ester of the free acid prostaglandin which corresponds to Bimatoprost. The free acid, 17-phenyl trinor PGF2α, is a potent FP receptor agonist.3 In human and animal models of glaucoma, FP receptor agonist activity corresponds very closely with intraocular hypotensive activity. The 17-phenyl trinor PGF2α isopropyl ester derivative is examined for IOP-lowering activity during the development of latanoprost.4 At the dose of 3 μg/eye in the monkey, 17-phenyl trinor PGF2α isopropyl ester is the most potent analog tested in reducing IOP, lowering the IOP 1.3 mm Hg below the level achieved by latanoprost. However, this derivative is also significantly more irritating to the eye than latanoprost.

Although much less potent that the parent compound, 15-keto PGs still retain the ability to produce a small but measurable decrease (1 mm Hg) in the intraocular pressure of normal cynomolgus monkeys when administered at a dose of 1 μg/eye.2 15-keto Latanoprost (15-keto-17-phenyl-13,14-dihydro trinor PGF2α isopropyl ester) is a miotic in the normal cat eye, causing an 8 mm reduction in pupillary diameter at 5 μg/eye. Again, this is not as potent as many other F-type PGs; for example, PGF2α will produce this degree of miosis at a dose of less than 1 μg/eye.

Semotiadil is a novel calcium antagonist whose stereochemical structure was investigated by X-ray crystallography and circular dichroism spectroscopy. X-ray structural analysis was performed using its enantiomeric salts with (S)-(+)-mandelic acid (2); 1 (mandelate salt). The R absolute configuration of 1, which was previously determined by chemical transformation of an optically active synthetic precursor, was directly confirmed by this analysis. Hydrogen bonding, charge repulsion, and hydrophobic interactions that stabilize the crystal structure were observed in the crystals. Circular dichroism spectra of 1 (mandelate salt) and 1 (hydrofuranate salt) in ethanolic solution showed similar spectral patterns. The CD bands of each chromophore were unambiguously assigned, and the conformation of the benzothiazole ring was determined using the optical rotation rule. The X-ray and CD data confirmed the P conformation of the benzothiazole ring and the equatorial position of the 2-phenyl ring in both the crystal and ethanolic solution states. These findings are in good agreement with our previous results on the conformational analysis of 2-arylbenzothiazole derivatives.

DCFBC is a prostate-specific membrane antigen (PSMA) inhibitor for small animal positron emission tomography (PET) imaging. [18F]DCFBC was prepared by reacting fluorine-18 labeled phenyl bromide with the precursor (S)-2-[3-[(R)-1-carboxy-2-thiolethyl]urea]-glutaric acid in ammonia-saturated methanol at 60°C for 10 min and then purified by C-18 reversed-phase HPLC. [18F]DCFBC was injected via the tail vein in severe combined immunodeficient mice for in vitro biodistribution or imaging. For in vitro biodistribution studies, mice were sacrificed at 5, 15, 30, 60, and 120 min after injection, and tumors, blood, and major organs were collected, weighed, and radioactivity was counted. Imaging was performed using a GE eXploreVista small animal PET scanner, collecting 12 consecutive 10-min frames. Results showed that the radiochemical yield of [18F]DCFBC averaged 16±6% (n=8) from 4-[18F]fluorophenyl bromide. Specific radioactivity ranged from 13 to 133 GBq/Amol (350-3600 Curie/mmol) with an average of 52 GBq/Amol (1392 Curie/mmol; n=6). Biodistribution and imaging studies showed high uptake of [18F]DCFBC in PIP tumors and almost no uptake in FLU tumors. High radiopharmaceutical uptake was also seen in the kidney and bladder; however, radioactivity washout from these organs was faster than from the PIP tumors. Maximum PIP tumor uptake was reached at 60 min post-injection at 8.16±2.55% injected dose/g and decreased to 4.69±0.89 at 120 min post-injection. The PIP tumor-to-muscle ratio was 20 at 120 min post-injection. Based on mouse biodistribution, the dose-limiting organ was the kidney (estimated human absorbed dose: 0.05 mGy/MBq; 0.2 rad/mCi). Conclusions: [18F]DCFBC localizes specifically to PSMA+ expressing tumors in mice and is suitable for small animal PET imaging. This novel radiopharmaceutical is an attractive candidate for further study in PET imaging of prostate cancer.

15(R)-17-phenyl trinor Prostaglandin F2α isopropyl ester (15(R)-17-phenyl trinor PGF2α isopropyl ester) is the latanoprost-related isomer containing both a double bond at 13,14 and an inverted (β) hydroxyl group at C-15. Similar to 15(S)-latanoprost, 15(R)-17-phenyl trinor PGF2α isopropyl ester is a potential impurity in most commercial preparations of the latanoprost bulk drug product. The IC50 values for the free acid forms of 15(S)-17-phenyl trinor PGF2α and 15(R)-17-phenyl trinor PGF2α were determined to be 0.71 nM and 30 nM, respectively, in a FP receptor binding assay using the cat iris sphincter muscle.1 A 3 μg dose of 15(R)-17-phenyl trinor PGF2α caused a 1.9 mmHg reduction of IOP in normotensive cynomolgus monkeys.
